Mechanisms of regulation of autophagy in response to sulfur starvation
Autophagy is a key lysosomal degradative and recycling process essential for cell survival during nutrient starvation. The yeast Saccharomyces cerevisiae has been a powerful and decisive model to investigate the genetic and molecular bases of autophagy and its regulation.
However, studies in yeast have mainly focused on nitrogen starvation, and little is known regarding other elemental nutrients. Our research focuses more specifically on sulfur, a vital element found notably in the amino acids methionine and cysteine, the ubiquitous methyl-group donor S-adenosylmethionine (SAM) and the antioxidant tripeptide glutathione (GSH).
We have discovered a new mechanism of induction of autophagy under sulfur starvation that depends on the transcription activator Met4 already known for its role in regulating sulfur metabolism genes (Prigent et al 2024). Our results indicate that cell survival under sulfur starvation relies on the interplay between sulfur metabolism and autophagy. We also show that SAM plays a key role in the signaling pathways that lead to the induction of autophagy under sulfur starvation. Following these findings, we are seeking to identify the actors involved in this pathway.

Mécanismes de régulation de l'autophagie en réponse à une carence en soufre
L’autophagie est un processus de dégradation lysosomale et de recyclage qui joue un rôle majeur dans la survie cellulaire en condition de carence nutritionnelle. La levure Saccharomyces cerevisiae a été un modèle déterminant pour l’étude des fondements génétiques et moléculaires de l’autophagie et de sa régulation.
Cependant la grande majorité des études réalisées à ce jour chez la levure utilise la carence en azote comme modèle de stress nutritionnel et les mécanismes de régulation de l’autophagie en réponse aux autres types de carence sont encore mal connues. Nous nous intéressons plus particulièrement à la carence en soufre, un élément vital présent notamment dans les acides aminés méthionine et cystéine, le donneur de groupe méthyle S-adénosylméthionine (SAM) et l'antioxydant glutathion (GSH).
Nos derniers travaux mettent en évidence en condition de carence en soufre un nouveau mécanisme d'induction de l'autophagie reposant sur l’activateur transcriptionnel Met4 déjà connu pour son rôle dans la régulation des gènes du métabolisme soufré (Prigent et al 2024). Nos résultats font apparaître que la survie cellulaire en carence en nutriments soufrés nécessite une coopération entre le métabolisme soufré et l’autophagie.
Nous montrons également que la SAM joue en carence en soufre un rôle primordial dans la signalisation aboutissant à l’induction de l’autophagie. Dans la suite de ces découvertes nous cherchons notamment à identifier les acteurs impliqués dans cette signalisation par la SAM.
