Study of bacterial nanomachines involved in antibiotic resistance and virulence using structural approaches

This presentation focuses on the structural study of type IV bacterial nanomachines involved in the dissemination of antibiotic resistance and virulence.
My work has led to the determination of several key structures of these systems using cryo-electron microscopy, revealing their assembly principles and functional mechanisms. More recently, I have developed an integrative approach combining microbiology, biochemistry, structural biology, and protein–protein interaction prediction to dissect the critical steps of bacterial conjugation, in particular recipient-cell engagement and substrate selection.
This research programme aims to establish a comprehensive mechanistic understanding of these nanomachines, with the long-term goal of identifying new strategies to limit the spread of antibiotic resistance.

Étude de nanomachines bactériennes impliquées dans la résistance aux antibiotiques et à la virulence par des approches structurales
Habilitation à diriger des recherches (HDR)
Cette présentation porte sur l’étude structurale des nanomachines bactériennes de type IV impliquées dans la dissémination de la résistance aux antibiotiques et la virulence.
Mes travaux ont permis de résoudre plusieurs structures clés de ces systèmes par cryo-microscopie électronique, révélant leurs principes d’assemblage et de fonctionnement. Plus récemment, j’ai développé une approche intégrative combinant microbiologie, biochimie, biologie structurale et prédiction d’interactions protéiques afin de décrypter les étapes critiques de la conjugaison bactérienne, notamment l’engagement avec la cellule receveuse et la sélection des substrats.
Ce programme vise à établir une compréhension mécanistique globale de ces nanomachines, avec pour objectif d’identifier de nouvelles stratégies pour limiter la propagation de la résistance aux antibiotiques.
