Repairing a broken chromosome

The repair of a broken chromosome by homologous recombination involves a complex series of steps whereby the broken ends are resected, bound by Rad51 recombinase and engaged in a search for a homologous DNA sequence with which it can pair and initiate new DNA synthesis to patch up the break. Using budding yeast, we have monitored many of the individual steps in this process, with a present emphasis on the search for homology, the fidelity of DNA synthesis, the maintenance of chromatin marks, and the cellular response to DNA damage.
