Bacterial conjugation is the unidirectional transport of genetic materials from a donor to a recipient cell. It is also the principal means by which antibiotic resistance genes spread among bacterial populations. It is mediated in the donor cell by three major complexes that work in concert: a DNA-processing machinery called the relaxosome, a double-membrane spanning transfer machinery termed type 4 secretion system (T4SS), and an extracellular appendage termed pilus.
While the structures of the T4SS and pilus are known, that of the relaxosome is not. Here, we describe the cryo-EM structure of a fully assembled relaxosome encoded by the F plasmid. This structure shed unique insights into a crucial step in the mechanism of bacterial conjugation, that of DNA processing and transfer.
Gabriel Waksman is professor of biochemistry and molecular biology at University College London (UCL) and professor of structural and molecular biology at Birkbeck College, University of London. His research focuses on the mechanisms of bacterial secretion, using mainly X-ray crystallography and electron microscopy.

As part of the partnership with the Life and Health Sciences doctoral school, participation in the ‘IGDR Keynote Lectures’ is included in the doctoral students' training hours at the rate of 1 hour/lecture, with a minimum attendance of three lectures.

La conjugaison bactérienne est le transport unidirectionnel de matériel génétique d'une cellule donneuse à une cellule receveuse. C'est également le principal moyen par lequel les gènes de résistance aux antibiotiques se propagent au sein des populations bactériennes. Dans la cellule donneuse, la conjugaison est assurée par trois grands complexes qui travaillent de concert : une machinerie de traitement de l'ADN appelée relaxosome, une machinerie de transfert à double membrane appelée système de sécrétion de type 4 (T4SS) et un appendice extracellulaire appelé pilus.
Si les structures du T4SS et du pilus sont connues, celle du relaxosome ne l'est pas. Nous décrivons ici la structure cryo-EM d'un relaxosome entièrement assemblé, codé par le plasmide F. Cette structure apporte un éclairage unique sur une étape cruciale du mécanisme de conjugaison bactérienne, celle du traitement et du transfert de l'ADN.
Professeur de biochimie et biologie moléculaire à l'University College London (UCL), Gabriel Waksman est également professeur de biologie structurelle et moléculaire au Birkbeck College, Université de Londres. Ses recherches portent sur les mécanismes de sécrétion bactérienne, en utilisant principalement la cristallographie aux rayons X et la microscopie électronique.

Dans le cadre du partenariat avec l’école doctorale Sciences de la Vie et de la Santé, la participation aux conférences "IGDR Keynote Lectures" est prise en compte dans les heures de formation des doctorantes et doctorants à raison de 1h00/conférence, avec une participation minimale à trois conférences.
